Inhibrx Combo Shows 44% Response vs 21% With Pembrolizumab Alone in First-Line Head and Neck Cancer

The early Phase 2 readout supports a move to Phase 3 in Q3 2026.

Overview

  • Inhibrx disclosed Monday interim Phase 2 results in first-line, PD-L1–positive head and neck cancer, and the stock rose about 17% after the release.
  • The combination of INBRX-106, an OX40-activating antibody, with pembrolizumab produced a 44% confirmed objective response rate compared with 21.4% for pembrolizumab alone.
  • Among evaluable patients, 11 of 25 responded on the combo versus 6 of 28 on monotherapy, and the combo yielded three complete responses while the control arm had none.
  • Blood analyses showed up to a 15-fold rise in CD8 and CD4 T-cell proliferation with the combo versus up to 2.5-fold with pembrolizumab alone, aligning with OX40’s role in boosting anti-tumor T cells.
  • Inhibrx reported a manageable safety profile with mostly mild rash, diarrhea, fatigue and infusion reactions, and it plans to begin Phase 3 in Q3 2026 with Phase 2 progression-free survival data expected in Q4 2026.
